  记者日前从中科院上海生科院获悉,该院生化与细胞所的科研人员阐明抑癌基因LKB1抑制肺癌转移的新机制,并发现赖氨酰氧化酶是肺癌转移治疗中潜在的分子靶点,这将为肺癌的临床治疗提供理论基础和新的策略。该成果日前在线发表在国际知名学术期刊《ProceedingsoftheNationalAcademyofSciences》上。
  肺癌是当今全球范围内危害性最大的疾病之一,具有高发病率和高致死率。超过90%的癌症患者死于肿瘤的转移和扩散。肺癌转移是一个复杂过程,其中癌细胞和细胞外基质的“亲密互动”是肿瘤转移过程中一个很重要的事件。科学家发现,肺癌中有一个新的抑癌基因LKB1,其功能性缺失突变在人类非小细胞肺癌中普遍高发,而且LKB1的功能丧失可以显著地促进肺癌的转移,但具体作用机理一直不清楚。
  中科院上海生科院生化与细胞所的季红斌课题组和葛高翔课题组通过合作,首次阐明LKB1的作用机制。研究发现,原来微环境中肿瘤转移的相关基因赖氨酰氧化酶LOX受LKB1的信号调控,在LKB1缺失的情况下,LOX会变得活跃,激活信号通路增强细胞的侵袭能力,成为肿瘤转移的“帮凶”。在肺癌小鼠模型中,通过抑制LOX的方法,可以显著减轻肺癌的发生和恶性转移。
